Synthesis and initial characterization of the ligand 6-((bis((4-methoxypyridin-2-yl)methyl)amino)methyl)picolinic acid (p-OMe TPAMA) are presented. This ligand consists of the same components as the previously studied 6-((bis(pyridin-2-ylmethyl)amino)methyl)picolinic acid (TPAMA), except with para methoxy substituents on two of the pyridine rings. The synthesis and initial characterization of the corresponding Mn(II) complex of p-OMe TPAMA is reported, and the complex¿s potential as an alternative MRI contrast agent analyzed. The pKa values for the ligand and its Mn(II) complex are determined via potentiometric titration, and the speciation determined through the fitting of the data. The resultant data suggests that the methoxy substituent decreases the stability of the complex. The effect of the substituent on the complex¿s relaxivity is determined through pH-dependent r1 relaxivity measurements. The r1 value at physiological pH was measured to be 1.56 mM-1sec-1, a decrease from that of the unsubstituted ligand.
